چکیده

Cationic polymers have been regarded as promising non-viral gene carriers because of their advantages over viral gene vectors, such as low cost, a high level of safety and easy manipulation. However, their poor transfection efficiency in the presence of serum and high toxicity are still limiting issues for clinical applications. In addition, the lack of adequate understanding of the gene delivery mechanism hinders their development to some extent. In this study, new polycations (PAPEs) consisting of a low generation polyamidoamine (PAMAM) core and branched polyethyleneimine (PEI-1.8k) outer layers were synthesized and their transfection activity and mechanism were studied. PAPEs were characterized by FTIR, (1)H NMR and gel permeation chromatography. PAPEs were able to self-assemble with pDNA and form spherical nanoparticles with sizes of 70-204 nm and zeta potentials of 13-33 mV. Importantly, the PAPE-pDNA complexes displayed lower cytotoxicity and higher transfection activity than PEI 25k in various cell lines, specifically in the presence of serum. The transfection mechanism was evaluated by endocytosis inhibition with specific inhibitors, time-dependent transfection, and intracellular trafficking inspection by CLSM. The high levels of transgene expression mediated by PAPEs were attributed to caveolae-mediated cellular uptake, the reduced entry into lysosomes and the entry into the nucleus through mitosis.

Modified Polyethylenimine: Self Assemble Nanoparticle Forming Polymer for pDNA Delivery

Objective Polyethylenimine (PEI), a readily available synthetic polycation which has high transfection efficiency owing to its buffering capacity was introduced for transfection a few years ago. But it has been reported that PEI is cytotoxic in many cell lines. In this study, in order to enhance the transfection efficiency of 10 kDa PEI and reduce its toxicity, hydrophobic residues were grafte...

Optimization of conditions for gene delivery system based on PEI

Objective(s): PEI based nanoparticle (NP) due to dual capabilities of proton sponge and DNA binding is known as powerful tool for nucleic acid delivery to cells. However, serious cytotoxicity and complicated conditions, which govern NPs properties and its interactions with cells practically, hindered achievement to high transfection efficiency. Here, we have tried to optimize the properties of ...

Fabrication, characterization and in vitro evaluation of poly(D,L-lactide-co-glycolide) microparticles loaded with polyamidoamine-plasmid DNA dendriplexes for applications in nonviral gene delivery.

We report, for the first time, on the preparation, characterization and in vitro testing of poly(D,L-lactide-co-glycolide) (PLGA) microparticles loaded with polyamidoamine (PAMAM)-plasmid DNA (pDNA) dendriplexes. Loading of pDNA into the PLGA microparticles increased by 150% when pDNA was first complexed with PAMAM dendrimers relative to loading of pDNA alone. Scanning electron microscopy (SEM)...

Biscarbamate cross-linked polyethylenimine derivative with low molecular weight, low cytotoxicity, and high efficiency for gene delivery

Polyethylenimine (PEI), especially PEI 25 kDa, has been widely studied for delivery of nucleic acid drugs both in vitro and in vivo. However, it lacks degradable linkages and is too toxic for therapeutic applications. Hence, low-molecular-weight PEI has been explored as an alternative to PEI 25 kDa. To reduce cytotoxicity and increase transfection efficiency, we designed and synthesized a novel...
